If you unlocked the Ultimate Lair previously will it automatically be available on NG+ or do you have to do the same number of side quests again?

You still have to complete a high number of side quests to unlock the extra day. I don't think it's like Last One Standing; most things are reset on a NG+, including side quest progression. On the plus side, side quests are much easier on a NG+ as you likely have most items for fetch/Wildlands quests already.
